Use the extraction buffer provided....

I have used these cards quite a bit. They come with a small vial of extraction buffer and a small disposable pipet. You can moisten a sterile swab in the buffer and roll the swab over the dried stain. Pop the swab back in the vial for a few minutes and then using the pipe transfer a few drops of the extract in the vial to the card. Alternatively, cut a small square from the stain and place it in the vial with the extraction buffer.

Hi, I believe you have to collect the blood with a sterile swab moistened with sterile water.
